Die Untersuchung mit der FISH-Methode wird derzeit in unserem Labor f\u00fcr das pr\u00e4implantationsgenetische Screening (PGS (PGT-A)) ausgew\u00e4hlter Chromosomen (in der Regel 5 der h\u00e4ufigsten ganzchromosomalen numerischen Abweichungen) und f\u00fcr die pr\u00e4implantationsgenetische Diagnostik\/Testung unbalancierter Formen famili\u00e4rer Umstrukturierungen (PGT-SR) kleinen Umfangs verwendet, bei denen die NGS-Methode nicht angewendet werden kann.<\/p>\n\n<p><strong>Prinzip der FISH-Methode:<\/strong> Untersucht werden Zellkerne, die auf einem Objekttr\u00e4ger fixiert sind. Das Prinzip der Methode besteht in der Markierung mehrerer spezifischer Chromosomenbereiche mit einer fluoreszenzmarkierten Sonde, die komplement\u00e4r zu dem jeweiligen DNA-Abschnitt ist. Die Auswertung erfolgt mittels Fluoreszenzmikroskopie. Mit der Methode k\u00f6nnen nur numerische Ver\u00e4nderungen ausgew\u00e4hlter Chromosomen bzw. ihrer Teile bestimmt werden, die mit der gew\u00e4hlten fluoreszierenden Sonde markiert sind. Bei der Durchf\u00fchrung von PGD (PGT-SR) zur Feststellung unbalancierter Formen famili\u00e4rer Chromosomenumstrukturierungen handelt es sich um Teile der Chromosomen, die an dieser Aberration beteiligt sind. PGD (PGT-SR) mit der FISH-Methode geht in der Regel eine Chromosomenuntersuchung der Eltern voraus, um die Genauigkeit der vorgeschlagenen Kombination von fluoreszierenden Sonden zu \u00fcberpr\u00fcfen (sogenanntes \u201eSet-up\u201c).<\/p>\n\n<p><strong>Einschr\u00e4nkungen der FISH-Methode:&nbsp;<\/strong>Mit FISH k\u00f6nnen keine weiteren Krankheiten oder Entwicklungsst\u00f6rungen des F\u00f6tus ausgeschlossen werden, die nicht durch eine Ver\u00e4nderung der Anzahl der untersuchten Chromosomenbereiche verursacht werden.<\/p>\n\n<h3>Schema der
